Resumo: Metabolomics is an analytical tool used to detect metabolites in biofluids or tissues and thereby identify and quantify metabolites commonly detected in the human serum metabolome. Cancer cells possess a unique metabolic phenotype that makes it possible to identify fingerprints, profiles or signatures to detect cancer and evaluate the pharmacodynamic effects of therapy. Krebs cycle acids have the potential to be used as biomarkers when used with other metabolites profile. Problems resulting from the analysis of fatty acids by LC-MS have been resolved with the use of N-(4-aminomethylphenyl) pyridinium (AMPP) derivatization. Through this project we show that it is possible to derivatize some of the Krebs acids and lactic acid using that same process used for fatty acids. We also proved that is possible to retain derivatized pyruvic acid under LC-MS conditions.
